An evaluation of the ICPL (isotope-coded protein labeling) non-isobaric labeling technique was performed using two different biological models. Two samples containing phage T4 capsids were mixed in a 1:1 ratio after being labeled with the light or heavy versions of the ICPL reagent. The analysis of this proteome demonstrated the feasibility of this approach for differential quantitative proteomics and was employed to optimize the experimental parameters of the ICPL workflow. ICPL-mediated analysis of two more complex proteomes, those of a Salmonella enterica serovar Typhimurium virulent strain and an isogenic attenuated mutant, and its comparison with the results obtained in a 2D-PAGE “classical” approach confirmed that ICPL is a valuable alternative to other labeling techniques currently in use. In addition, our results suggest that labeling at the peptide level instead of following the standard ICPL workflow should increase both the number of proteins quantified and the reliability of the quantification. 相似文献

This paper presents a finite element algorithm for the simulation of thermo‐hydrodynamic instabilities causing manufacturing defects in injection molding of plastic and metal powder. Mold‐filling parameters determine the flow pattern during filling, which in turn influences the quality of the final part. Insufficiently, well‐controlled operating conditions may generate inhomogeneities, empty spaces or unusable parts. An understanding of the flow behavior will enable manufacturers to reduce or even eliminate defects and improve their competitiveness. This work presents a rigorous study using numerical simulation and sensitivity analysis. The problem is modeled by the Navier–Stokes equations, the energy equation and a generalized Newtonian viscosity model. The solution algorithm is applied to a simple flow in a symmetrical gate geometry. This problem exhibits both symmetrical and non‐symmetrical solutions depending on the values taken by flow parameters. Under particular combinations of operating conditions, the flow was stable and symmetric, while some other combinations leading to large thermally induced viscosity gradients produce unstable and asymmetric flow. Based on the numerical results, a stability chart of the flow was established, identifying the boundaries between regions of stable and unstable flow in terms of the Graetz number (ratio of thermal conduction time to the convection time scale) and B, a dimensionless ratio indicating the sensitivity of viscosity to temperature changes. Sensitivities with respect to flow parameters are then computed using the continuous sensitivity equations method. We demonstrate that sensitivities are able to detect the transition between the stable and unstable flow regimes and correctly indicate how parameters should change in order to increase the stability of the flow. This paper focuses on modelling the severity distribution. We directly model the small, moderate and large losses with the Pareto Positive Stable (PPS) distribution and thus it is not necessary to fix a threshold for the tail behaviour. Estimation with the method of moments is straightforward. Properties, graphical tests and expressions for value-at risk and tail value-at-risk are presented. Furthermore, we show that the PPS distribution can be used to construct a statistical test for the Pareto distribution and to determine the threshold for the Pareto shape if required. An application to loss data is presented. We conclude that the PPS distribution can perform better than commonly used distributions when modelling a single loss distribution for moderate and large losses. This approach avoids the pitfalls of cut-off selection and it is very simple to implement for quantitative risk analysis. 相似文献

In this paper, a robust projection method on a locally refined mesh is proposed for two‐ and three‐dimensional viscous incompressible flows. The proposed method is robust not only when the interface between two meshes is located in a smooth flow region but also when the interface is located in a flow region with large gradients and/or strong unsteadiness. In numerical simulations, a locally refined mesh saves many grid points in regions of relatively small gradients compared with a uniform mesh. For efficiency and ease of implementation, we consider a two‐level blocked structure, for which both of the coarse and fine meshes are uniform Cartesian ones individually. Unfortunately, the introduction of the two‐level blocked mesh results in an important but difficult issue: coupling of the coarse and fine meshes. In this paper, by properly addressing the issue of the coupling, we propose a stable and accurate projection method on a locally refined staggered mesh for both two‐ and three‐dimensional viscous incompressible flows. The proposed projection method is based on two principles: the linear interpolation technique and the consistent discretization of both sides of the pressure Poisson equation. The proposed algorithm is straightforward owing to the linear interpolation technique, is stable and accurate, is easy to extend from two‐ to three‐dimensional flows, and is valid even when flows with large gradients cross the interface between the two meshes. The resulting pressure Poisson equation is non‐symmetric on a locally refined mesh. The numerical results for a series of exact solutions for 2D and 3D viscous incompressible flows verify the stability and accuracy of the proposed projection method. The method is also applied to some challenging problems, including turbulent flows around particles, flows induced by impulsively started/stopped particles, and flows induced by particles near solid walls, to test the stability and accuracy. In this paper, the pattern memory of the second-order neural networks with time-delay are investigated based on the stability theory. Several sufficient conditions are obtained such that the equilibrium point is locally exponential stable when the equilibrium point located in the saturation region. These conditions can be directly derived from the synaptic connection weights and the external input of the second-order neural networks, and be verified easily.
